For me it's the upcoming Hawkman minifigure, simply because he's one of my favourite comic book characters but the desirability is raised because the previous approach Lego had taken to the DC superheroes theme left me resigned to the fact that I'd most likely never see an official one made. Add to that the fact that the rest of the Heroes in the wave are all Justice League mainstay characters, Hawkman being on the roster was a little bit of a surprise...he's often on the Justice League, but normally as a secondary member or in the current continuity he's part of a different Justice League.
There are all sorts for me. I really like the older, more minimalist minifigs from the 'Classic' themes, but then again, I do like a really well-printed (ie, nice design, on torso, back, legs, face, etc) modern fig from licensed themes. And there's always a couple of cute CMFs per theme.
I think it's mostly if they have some sort of significance to me. I like 'Classic' minifigs (ie, classic space, castle) for the nostalgia factor - not that I was old enough to have owned any new, but I had quite a few second-hand sets. I like the Star Wars figs because they remind me of the fun times I had watching Clone Wars with my brother, playing Battlefront II and Republic Commando with my friends and so on. Likewise CMFs if they remind me of a movie/book/video game character.
I guess what I'm saying is that for me, exclusivity doesn't matter so much. You can keep your solid gold 3POs and SDCC Azogs.
All in all, that's not too helpful, is it? I haven't given any specifics. ;-)
Forestman - Pouch, Green Hat, Yellow - why they didn't include the pouch on the CMF one I can't imagine. I also liked the torso from the peasant in #6055 but the rest of the figure was pretty awful.
The original Forestwoman, too, has to be on the list given it's one of the first 'proper' female minifigs as opposed to one with a classic smiley.
Today it's figs, tomorrow it'll be "Now please tell us what sets will be worth money when they retire..." :oP lol.
Joking aside, I can't help but feel this is just milking/mining the lego fanbase now, without actually providing it/us with anything of actual substance in return. The very question is literally just a forum thread.
I don't mean to sound so negative about a forthcoming lego book, as I LOVE a good book, especially one about my favourite hobby... but 'good' book is the operative word. If they must print endless lego books, but are so short of ideas or insight, they really should ask us outright what book to publish. Otherwise i guess we can look forward to more threads lifted from forums masquerading as 'interest' books.
Blocks magazine came under fire for regurgitating the web. How is this going to be any different? ...(They'll stick a fig on the cover). *rolls eyes*

Firstly, if we take into account rarity, Mr Gold is my most valuable minifigure, followed closely by the SDCC Super Heroes exclusives. I enjoy owning rare minifigures in the knowledge that lots of people do not own them, but none of those are even close to my favourite minifigure. Only Phoenix and Bizarro are really of any use to me as I like those characters and would include them in a MOC if the opportunity arose.
The minifigure which conjures the fondest memories for me is probably the original Geonosian. #4478 was the first set I bought on the aftermarket back in 2006, and that one minifigure really got me focused on Star Wars as I loved flying it around the house.
When I was younger, and even now occasionally, I like to bring a minifigure when I am going for a long trip in the car. I was always thought that if the printing was too complicated it might rub off in my pocket (although this is highly unlikely as I realise now), and therefore I often brought along Captain Brickbeard from #6243 Brickbeard's Bounty. Somehow he always seemed unusual enough to keep me interested, but simple enough that I would not lose any pieces or damage him. In terms of memories, again this is a favourite.
For modern minifigures which I like due to detailed printing or simply because I like character, I have two contenders. The first of these is Darth Malgus, who is a brilliant character in the Star Wars: The Old Republic trailers and is translated perfectly into a minifigure. The other is the new Snowtrooper for 2014. I love Snowtroopers and to see a truly definitive version of such a great army builder minifigure is a treat.

I am a huge Star Wars fan and you can imagine my delight when I heard that my favorite movie franchise was going to be immortalized in my favorite building blocks. The day I held a Lego Darth Vader in my hand was a day I will remember forever :)
I am a Collectible Minifigure collector, so I desire a Mr. Gold because I don't consider my collection complete without him.
My desire is to have every Star Wars Minifigure ever made including the Gold C-3PO and the Bronze Boba Fett :)
My favorite Minifigures are the Classic Spacemen, Darth Vader, New Boba Fett, CMF Spartan and Zombie.

If I really wanted one, I could just stump up the cash on bricklink or ebay. But the fun is in hoping to find a bargain one day.
I'm thinking particularly the large amount of discussion that goes on in just about every Lego forum regarding the minifigures that have not been 'missed' from licenced themes like LOTR/Hobbit or Superheroes.
As a minifig collector, I've gotta say the original Lego spacemen. For a collector, they're as common as dirt, but finding ones with original thin-strap helmets and unfaded gold is almost impossible.
Other people it's bound to be the purists that want a real Mr gold, c3po, or SDCC figures that cost probably at least couple hundred each on the aftermarket instead of just buying a bootleg or custom for a fraction of the price.
For me, the figures I most want are the ones with PLAIN torsos that have a lot more value in my collection compared to a lot of the printed torsos out there. The red bob torso is a personal favorite and I have 4 in use in my collection at the moment, one or two spares walking around, and more bob torsos on my wish list along with 4 other colors.
If its the latter, my favourite include Loki, Iron Man, main LOTR/Hobbit minifigs such as Erlond, Arwen, Galadriel, dwawes etc, most of the latest HP ones such as Weasley family, Hagrid, other professors and students, Russian lady from Indiana Jones (I think one of the coolest female minifigs out there!), some of the stormtroopers, Greedo, Luke and Leia Skywalker, mummy and some monsters from Monster Fighters... There are more, including those I haven't come across yet! I think for me the criteria is previous connection with the character, and then the quality of the minifigure itself, how well it replicates the character and how good the minifigure looks.
Looking back on when I first got into Lego I would have to say that my favorite mining was the 2004 Greedo. I remember going over to my neighbors house who had this minifig and drooling over it. After many months of ogling it I finally trades another mini fig for it. I remember thinking how cool the printing, and how rare it must be. Of course know I know it isn't the most special mini fig in the world be at the time it was to me
So for me, it will be unsurprisingly my Jango Fett minifigure(s). There's a bit of a story in it too. Back when I was not much of a LEGO collector, about 2003 or so, I was in Kb toys (rest in peace). I noticed Jango Fett's Slave 1 on a shelf marked at 50% off. Jango Fett was one of my favorites from the Star Wars movies so I picked it up thinking what was the harm in a small $25 buy. I built it and thought it was incredible, yet still didn't think much of LEGO as a hobby. It got put on shelf (and broke multiple times due to being bumped off the shelf). Somehow, the minifigures got separated and as a prank Jango Fett was hidden in a VCR box (the person that hid it forgot they hid it). I completely forgot about Jango Fett until 2 years later when I was searching through eBay and noticed that Jango Fett was commanding prices of $100+. I anxiously searched the house (hoping for the quick buck), but had no luck. Several months later, a young relative wanting to watch Veggie-Tales found a LEGO minifigure hidden in the back of the box. Lo and behold it was Jango Fett! I immediately listed and sold it. With my now $100 I decided that I wanted to buy... more LEGO. Thus, the hobby began and everything in my collection has actually grown out of that $100 I got from Jango Fett. I've never really put another dime into it. Self funding hobby indeed!

For me $$$ value is of no consequence, mr gold might be worth lots but i woukd never display it - its a nasty little tacky trinket with almost zero design value. However I can see that a collector would see it completely differently.

I'm easy to please, I have my 100 most desired minifigs (and more).
Of course, if financial matters come into it, then I desire the top 100 most expensive. For the simple reason that I would sell them and buy 1000 of what I actually prefer.
There could be details on the rarity of each, where figures are easy to source, where arms and legs don't need detaching from their torsos and hips, etc.
Could be a nice little featurette to break up the sections. I'm not into them yet, but I like seeing that sort of info about them and I wonder how long it'll be till I start trying to get some together.

My favorite minifigure in my collection is sw547 Darth Revan, since I never thought in a million years they'd make an official one. He's iconic to me after spending an entire summer playing KOTOR. Really well done minifigure. Detailed but no too detailed like a lot of new SW minifigs are. I'm so glad he was a freebie with purchase rather than a con exclusive.
I also like the semi-humanoid sea-creatures from the Atlantis series, the seashell-encrusted monsters from Pirates of the Caribbean, the winged Geonosians from Star Wars, and almost all the figures from the Lord of the Rings.
And I enjoy collecting the statues...the granite Classic Space statue being my favorite!
I am surprised about the POC. I think they are like batman and did not sell that well.
I'd love to own Mr Gold from the series 10 CMF, this is because I need him to complete my collection. The other minifigures that I would love to own are the ones that are available at comic conventions however due to their high selling point this will never happen (unless I win the lottery).
The minifigures that are my favourite from my collection
One of my most favourite minifigures from my collection is the Heath Ledger Joker from the tumbler set as he is original to the film character. The other minifigures that are worthy of a mention are: Delores Umbridge from the Hogwarts castle (as she is exclusive to that set) and her printing is fantastic). Rocket racoon from the GOTG set is also another favourite of mine. I do have plenty more but these are my favourites. :-D

I just said some because I had read several list that spoke of rarity, but it seemed more like a price list. NYTF iron man and cap were lower than mr gold. That's not right if it's a rarity list
More realistically Deadpool and Wolverine are great, Catwoman is good, and the Joker with the Tumbler is cool too. I'd quite like a Harley Quinn figure as well, I missed all the cheaper sets with them, maybe they will bring one out again in the future.
Robot Emmet is great, hopefully they will do a matching Robot Wildstyle soon as well.

All the historical, mythological, and fantasy minifigs are definitely amongst my favourites.
Furthermore I agree with @Drmnez
Not only do I like the Medusa minifig a lot, I bought an entire CMF series 10 box, and there were only two Medusa minifigs in it. So compared to other minifigs from that series Medusa was produced in much smaller numbers too.
Funny that you mention that, because back when I opened the CMF bag containing Medusa, the first thing I tried was to figure out if and how a minifig could hold her head and hair piece by one of the snakes in his hand. And although the snake-hairpiece does not have a minifig hand/clutch 'handle', on purpose or by accident a minifig can hold up medusa's head by holding on to the top right snake tail (which is on the left upper side of the photo of the CMF medusa). In my opinion that works quite well for re-enacting, or creating a diorama of the scene where Perseus confronts and turns the Kraken into stone. Especially when you have a minifig hold the head and hairpiece with its right hand so the minifig stands facing somewhat sideways, position his head/face averted a little extra to the left and voila! He won't turn into stone himself. Also the hairpiece is made of the rubbery stuff so it will stick in a minifig's hand quite snug.
I think the Classic Town, Space and Castle minifigures would top my list. Most of them have been out of production for over 25 years or longer, so this are truly rare. In the case of Castle and Space there have been no figures produced that like the Classic ones.
I start with Castle as I'm a Castle collector and know it best. The Classic knights of 375/6075 and 383/6083 are unlike nearly anything else Lego has ever made. Between the visors, torsos and the stickers there are rare and definitely some of the hardest to find Castle minifigures.
Classic Spacemen are highly desirable. The subtheme's enduring popularity is as strong as ever. And the inclusion in the Lego Movie is another boost. The blue and black are the two most desirable Classis Spacemen. Blue for Benny and black for the rarity.
Until recently I would have say the Maersk worker was the top Classic minifigure. The release of the new Maersk train has had a huge impact on this, and I'm not sure what the current view on the original is. The girl with the white pigtails is rare and always seems to be in demand. The other ones that I know are rare and in high demand are the Guards from 1592 and the sticker version of the Exxon truck driver.
