Best Series: Classic LEGO Space
Best Set: #6970 Beta I Command Base
The Beta I Command Base had everything you needed. It had a cool spaceship, a great base, plenty of communication devices, a buggy, lots of spacemen, two crater plates *and* a monorail! Perfect.
All IMHO, of course!
Also cmfs, the Tower Bridge, the Sydney Opera House, the Taj Mahal (that I sadly missed but am currently bricklinking) and other landmarks that are to come.
Edit: Oh, and also everything that has a lot of pieces! :o)

Series: Space Police I
Favourite series would have to be Monster Fighters or possibly LOTR.
Favorite Theme: Probably have to go with Creator 3in1. I try to avoid licensed / minifigure driven / sticker heavy sets. So, 3in1 and the recent Modulars are where its at for me.
Favourite set : Portal of Atlantis. at least until I build tower of orthanc. well, I at least hope tower of orthanc is better for the price it cost.
The favourite set I own would have to be the tumbler just because I think it looks wonderful.
Saying that, the build I am most looking forward to doing is my bricklinked version of 10179.

Favorite Theme: Creator (WV, Modulars, Trains, Planes and Automobiles)
I have an affinity for the purple box Pirates, the Fire Ninjas, Forestmen and LotR.
And keychains.
If I think back to what I thought was "best" during my childhood, the answer is really "whatever I had". I had space sets because that was what was bought for me at the time but I didn't often play "space" with them.
Favorite themes:
1. Pirates 1989-1992
2. Castle 1988-1995
My favorite set would be Tower of Orthanc... if not for the stickers. Still waiting for that one favorite, I think. From my childhood, this would definitely be 5590: Whirl and Wheel Super Truck.
Theme? Creator Expert.
As for my favorite line, Creator by a mile.
Prior to last night, my favorite set was the 10188 Death Star. Not only was it a lot of fun building, but it only cost me $56 at a thrift store, with the original boxes and a bunch of spare parts from the 7893 Passenger Plane. And it ended the dark age.
My favorite set now is 3831 Rocket Ride from the Spongebob Squarepants theme. I haven't even finished finding all the parts, just got most of the rocket assembled, but my grandson spotted it last night and spent a solid hour showing me that it opens up and he can hide all manner of things inside it. He's also a big fan of the 42284/42291 Rocks.
the creator winter village theme comes a close second for me, love that line.
I don't know if I really have a favorite set but if I had to chose I would probably pick #70728 Battle for Ninjago City. It may get knocked out by #70751 Temple of Airjitzu which I'm in the process of building now.
#4559 Cargo Railway has a special place in my heart because it's the first train I ever bought and years after I sold the original one I had, I bought it again.
#6769-1 Fort Legoredo is another of the first sets I had bought about the same time that I bought that train. I wish I still had a complete one but I think I sold pieces of it, although I have the figures and horses.
Other themes up there would be Ninjago, any holiday theme lines (Winter Village, Monster Fighter, Scooby) and Modulars.
I think I also need to include Friends. It really has been a line that has broken the mould and just tried a wide variety of items never seen in Lego before.
Best set would probably be HH.
