Just wondering, is it possible to search in what I have in my wanted list by the following:
Total products in my list from a single shop +
Cheapest including shipping? (by total products + shipping)
if you guys know what I mean?

Also, what does Lots (Unique) mean in bricklink ?
BrickWizard and a matlab script.
A vendor may have several lot of the same brick, for example:
10 New Black 1x2 Bricks
12 Used Black 1x2 Bricks (like new)
15 Used Black 1x2 Bricks (scratched)
That is 3 Lots but 1 unique
If you don't mind splitting orders for less quantities, you could remove the quantity in your wanted list (but put it in the remark for you reference) or create a 2nd entry of the same with no min quantity.
Another useful tool to deal with Wanted List is BrickStock (desktop app), I have Bricklinked 25 parts model to 6000 parts model with just BrickStock and the Wanted List feature, could not have done it (so easily) without BrickStock as it allow you to manipulate lists and import them into BL. You can also put a price on each brick (or use the default average), so without a cost optimization program you can still see if a store has good prices.

Only used it a little bit but seems to work pretty well for me! Also doesn't include shipping cost, but you can narrow down the country search.
I was playing around with that matlab script, got it working in Oracle after some fiddling, but later an update to part of matlab that was needed for a missing feature broke it's Oracle compatibility. Hadn't heard of Brick Wizard before though.
