I'd like Patsy Walker, of course. Both Hawkeyes. Spidey 2099 is a current book, so I'd expect to see that variant in a forthcoming Spidey set.
Spider-Gwen or a civilian Gwen Stacy is a must. The cruel part of me laments Gwen's exclusion in last year's bridge set.
Gwenpool for a total left field enthusiast set, with a reissued MODOK and a new Batroc.
The new Champions (Ms. Marvel, Nova, Spider-Man, Viv Vision, Cho Hulk, and Cyclops) would make for a great Spidey set. Re-issue Miles Spidey, Nova, and Ms. Marvel with standard arms, with the characters who haven't had minifigs.
The X-Men would be my big desire - in an ideal world it's the 80's Outback lineup or 90's team, but I'd take anything. A proper X-23 as Wolverine would be ace, with hair and a new dual molded Wolvie mask with long hair behind.
The Red Hood in the Batman CMF gives me hope that Mysterio is close.

^ With the tone of the Netflix series for Daredevil & Luke Cage being decidedly 'adult' (especially how Punisher is represented as not worrying about killing people) I think the chances of TLG producing these now is quite low. As for New Warriors - I couldn't remember what their line-up was before Googling: now that I have I'd like Firestar, Justice and Nova.

I'd be surprised if we didn't get a Daredevil team up in the Spidey sets at some point.
The new cartoon is to debut this summer or fall, and I expect sets to mine that for inspiration like they did Ultimate Spider-Man. Hopefully this means Spider-Gwen and and a return of Miles.
I really don't think punisher is fully out of the realm of possibility as he did appear in the Lego Marvel Superheros video game, but yeah, the show would kind of be an issue. 2099 Spider-Man and/or Noir Spider-Man would be awesome too.

May as well add Miss Martian to that. I can see her with red Sabine hair, or that hair that came with the CMF Cyborg girl (has that hair been used elsewhere?).
Zatanna would be fantastic, especially if we could get a new top hat/hair mold. Throw her in set with Etrigan (Minifig would be ideal, but I can see a Bigfig) and Batman to move it.
Kate Kane as Batwoman just like she appears in LEGO Batman 3.

DC: Deadman, Bigfig Doomsday, Hugo Strange, Shadowpact (Group), Ares, Hades, The Doom Patrol (Group), and a Reverse Flash. As a side note, I think it would be great if we got a battle pack for each Lantern Corp, with one major member in each.
Marvel: Punisher (Unlikely), Mephisto (Even more unlikely), Original Guardians of the Galaxy (Vance Astro, etc), Dormammu, Silver Surfer, Bullseye, and more X-Men.
I'd like more variety in buildings for DC Heroes sets.
I don't need another Arkham or Batcave.

The Sanctum Santorum, Airport, and Avengers Tower are my favourite sets because I'm tired of stockpiling Quinjets and cars. Daily Bugle was unimpressive, though.
Anyway, to keep the fantasy thought exercise rolling, I'd love to see Cyborg Superman and Star Sapphire pop up somehow. Also Mera.
Obviously it wouldn't be big enough to include all those figures but some of them would definitely be welcome.
I also wouldn't mind a Batman variant of Batman as a Green Lantern (if they think it'll move the set) since he has worn the ring several times and has an Elseworld story about it.

Also, a Kate Spencer Manhunter would be amazing since we mentioned them.

But since it detected the remnants of a Green Lantern ring (that Hal gave him to use) and his will to not be reprogrammed it left.

OK. I liked all the black lanterns but Spectre was the funniest.
I copied someone else's idea for the Tick's head but I need to try and do something to make his features stand out more. Lousy phone camera doesn't help with that either
It even presents itself with a car, and they love slapping cars into superhero sets.
